An unconverted try to Shane Rodney gave the Panthers a 4-0 lead after six minutes.
Whilst Manly took the lead soon after, a converted try to Preston Campbell ensured that the home team held the lead at the break, up 10-6.
The visitors regained the lead midway through the second half, but with less than two minutes remaining a Paul Franze kick into the in goal was grounded by Luke Priddis, with Penrith stealing victory at the death.
